RESUMO

In this work, a covalent organic framework (TADM-COF) with high crystallinity and large specific surface area (2597 m2 g-1) has been successfully synthesized using 1,3,5-(4-aminophenyl) benzene (TAPB) and 2,5-dimethoxy-p-phenyldiformaldehyde (DMTP). The COF was grown in situ on oxide particles to form core-shell nanocomposites (SiO2@TADM COF, Fe3O4@TADM COF and Co3O4@TADM COF) to realize its function as a shell material. Among them, the Co3O4@TADM COF with the highest electrochemical response to purine bases was further cross-linked with multi-walled carbon nanotubes (MWCNT) to construct a novel electrochemical sensor (Co3O4@TADM COF/MWCNT/GCE) for detection of purine bases. In this nanocomposite, Co3O4 possesses rich catalytic active sites, MWCNT ensures superior electrical conductivity and COF provides a stable environment for electrocatalytic reactions as the shell. At the same time, regular pore structure of the COFs also offers smooth channels for the transfer of analytes to the catalytic site. The synergistic effect among the three components showed remarkable sensing performance for the simultaneous detection of guanine (G) and adenine (A) with a wide linear range of 0.6-180 µM and low limits of detection (LODs) of 0.020 µM for G and 0.024 µM for A (S/N = 3), respectively. The developed sensor platform was also successfully applied in the detection of purine bases in thermally denatured herring DNA extract. The work provided a general strategy for amplifying signal of COF and its composite in the electrochemical sensing.

RESUMO

It is of great significance to propose simple methods to detect DNA bases sensitively for biological analysis and medical diagnosis. Herein, a highly crystalline polyimide covalent organic framework (TAPM-COF) has been successfully synthesized via a solvothermal route using pyromellitic dianhydride (PMDA) and tris(4-aminophenyl) amine (TAPA), which possessed large specific surface area (2286 m2 g-1) and excellent thermal stability. Intriguingly, the crystallinity of the TAPM-COF improved significantly with the increase of water content in the reaction medium. To verify this phenomenon, we synthesized TPPM-COF with two pores by pyromellitic dianhydride (PMDA) and N,N,N',N'-tetrakis(4-aminophenyl)-1,4-benzenediamine (TPDA), which bonding was similar to TAPM-COF. Furthermore, the prepared TAPM-COF-0.3 was used to construct a novel and independent electrochemical biosensor on glassy carbon electrode for simultaneously determination of adenine (A) and guanine (G) without other additives. However, to further improve signal of TAPM-COF in electrochemical sensing, the crystalline TAPM-COF-0.3 can be readily integrated with amino-functionalized multiwalled carbon nanotubes (NH2-MWCNT) to form core-shell TAPM-COF-0.3@NH2-MWCNT driven by a π-π stacking interaction for more sensitive electrochemical sensing toward purine bases. In comparison to TAPM-COF/GCE, the TAPM-COF@NH2-MWCNT/GCE exhibited more favorable linear range and lower limit of detection. The work provided a new strategy for amplifying signal of COF in the field of electrochemical sensors.

RESUMO

The vinyl chloride monomer (VCM), a common raw material in the plastics industry, is one of the environmental pollutants to which humans are mostly exposed. Thiodiglycolic acid (TDGA) in human urine is a specific biomarker of its exposure. TDGA plays an important role in understanding the relationships between exposure to the VCM and the identification of subgroups that are at increased risk for disease diagnosis. Therefore, its detection is of great significance. Here, we designed and established a ratiometric fluorescent sensor for TDGA by using Eu3+ as a bridge connecting the covalent organic framework (COF) and the energy donor molecule 2,6-dipicolinic acid (DPA) and named it DPA/Eu@PY-DHPB-COF-COOH. The sensor not only possesses the advantages of a ratiometric fluorescent sensor that can provide built-in self-calibration to correct a variety of target-independent factors but also presents high selectivity and high sensitivity. Currently, there are only a few reports on the detection of TDGA, and to the extent of our knowledge, this report is the first work on the detection of TDGA based on a COF system; so, it has an important reference value and lays a solid foundation for designing advanced sensors of TDGA.

RESUMO

The development of a simple and sensitive electrochemical sensing platform for levofloxacin (LVF) analysis is of great significance to human health. In this work, a covalent organic framework (TP-COF) was in situ grown on the surface of Sn-MoC nanospheres with nanoflower-like morphology through a one-pot method to obtain the TP-COF@Sn-MoC composite. The prepared composite was used to modify a glassy carbon electrode (GCE) to realize the sensitive detection of levofloxacin. TP-COF was formed by polycondensation of 2,4,6-tris(4-aminophenyl)-1,3,5-triazine (TAPT) and pyromellitic dianhydride (PMDA), in which C = O and C = N groups served as double active centers for the recognition and electrocatalytic oxidation of the target molecule. Meanwhile, the introduction of Sn-MoC improved the conductivity of the electrode. The TP-COF@Sn-MoC composite produced a strong synergistic effect and showed a high electrocatalytic ability toward levofloxacin oxidation. The linear range of LVF was 0.6-1000 µM and the limit of detection (LOD) was 0.029 µM (S/N = 3). In addition, the sensor has been successfully applied for the analysis of LVF in human urine and blood serum samples with acceptable recovery rates, demonstrating that the sensor was promising in practical applications.

RESUMO

Introduction: Prolonged exposure of train drivers to thermal discomfort can lead to occupational safety and health (OSH) risks, causing physical and mental injuries. Traditional method of treating human skin as a wall surface fail to observe accurate skin temperature changes or obtain human thermal comfort that adapts to the thermal environment. Methods: This study employs the Stolwijk human thermal regulation model to investigate and optimize the thermal comfort of train drivers. To minimize the time-consuming design optimization, a pointer optimization algorithm based on radial basis function (RBF) approximation was utilized to optimize the train cab ventilation system design and enhance drivers' thermal comfort. The train driver thermal comfort model was developed using Star-CCM+ and 60 operating conditions were sampled using an Optimal Latin Hypercube Design (Opt LHD). Results and Discussion: We analyzed the effects of air supply temperature, air supply volume, air supply angle, solar radiation intensity and solar altitude angle on the local thermal sensation vote (LTSV) and overall thermal sensation vote (OTSV) of the train driver. Finally, the study obtained the optimal air supply parameters for the Heating Ventilation and Air Conditioning (HVAC) in the train cabin under extreme summer conditions, effectively improving the thermal comfort of the driver.

RESUMO

The aim of this study was to improve the entrepreneurial performance (EP) and sports morality of college teacher-and-student entrepreneurs (i.e., college entrepreneurs). Consequently, psychological capital (PsyCap) is creatively combined with social and political education (SPE) to explore college entrepreneurs' EP and sports morality. First, following a theoretical model implementation, this article proposes several hypotheses. Then, a questionnaire survey (QS) was designed, and the data were analyzed. The results show that (1) gender has little impact on EP and sports morality; (2) PsyCap significantly affects the EP of college entrepreneurs at the age of 33-38 years; (3) in terms of educational background, average scores of PsyCap + SPE of bachelors are the highest, followed by masters or above, and finally, the college undergraduate; (4) the average score of PsyCap + SPE of married respondents is 4.0, while that of the unmarried is 3.7; (5) there is a significant difference between college entrepreneurs' EP under the dimension of the basic enterprise situation; and (6) the average score of the length of entrepreneurship is 9.87, which has the most significant impact on the EP and sports morality, and the significance of sports morality is 0.04. Among them, the most significant impact on EP and sports morality is weekly sports participation, with a score of 9.67. Therefore, PsyCap + SPE plays a positive role in the EP of college entrepreneurs. In contrast, sports morality has little impact on EP. This study provides a reference for the research on the influence of entrepreneurship and sports morality of college entrepreneurs.

RESUMO

In Arabidopsis thaliana, mitochondrial-localized heat-shock cognate protein 70-1 (mtHSC70-1) plays an important role in vegetativegrowth. However, whether mtHSC70-1 affects reproductive growth remains unknown. Here, we found that the mtHSC70-1 gene was expressed in the provascular cells of the embryo proper from the early heart stage onward during embryogenesis. Phenotypic analyses of mthsc70-1 mutants revealed that mtHSC70 deficiency leads to defective embryo development and that this effect is mediated by auxin. In addition to a dwarf phenotype, the mthsc70-1 mutant displayed defects in flower morphology, anther development, and embryogenesis. At early developmental stages, the mthsc70-1 embryos exhibited abnormal cell divisions in both embryo proper and suspensor cells. From heart stage onward, they displayed an abnormal shape such as with no or very small cotyledon protrusions, had aberrant number of cotyledons, or were twisted. These embryo defects were associated with reduced or ectopic expression of auxin responsive reporter DR5rev:GFP. Consistently, the expression of auxin biosynthesis and polar auxin transport genes were markedly altered in mthsc70-1. On the other hand, mitochondrial retrograde regulation (MRR) was enhanced in mthsc70-1. Treatment of wild-type plants with an inhibitor that activates mitochondrial retrograde signaling reduced the expression level of auxin biosynthesis and polar auxin transport genes and induced phenotypes similar to those of mthsc70-1. Taken together, our data reveal that loss of function of mtHSC70-1 induces MRR, which inhibits auxin biosynthesis and polar auxin transport, leading to abnormal auxin gradients and defective embryo development.

RESUMO

Background: The Coronavirus Disease 2019 (COVID-19) pandemic has led to overwhelming levels of distress as it spread rapidly from Wuhan, Hubei province to other regions in China. To contain the transmission of COVID-19, China has executed strict lockdown and quarantine policies, particularly in provinces with the highest severity (i.e., Hubei). Although the challenges faced by individuals across provinces may share some similarities, it remains unknown as to whether and how the severity of COVID-19 is related to elevation in depression. Methods: The present study compared depression among individuals who lived in mildly, moderately, and severely impacted provinces in China following the lockdown (N = 1,200) to norm data obtained from a representative sample within the same provinces in 2016 (N = 950), and examined demographic correlates of depression in 2020. Results: Residents in 2020, particularly those living in more heavily impacted provinces, reported increased levels of depression than the 2016 sample. Subsequent analyses of sub-dimensions of depression replicated the findings for depressed mood but not for positive affect, as the latter only declined among residents in the most severely impacted area. Increased depressed mood was associated with female, younger age, fewer years of education, and being furloughed from work, whereas reduced positive affect was associated with younger age and fewer years of education only. Conclusions: This study underscored the impact of COVID-19 on depression and suggested individual characteristics that may warrant attention.

RESUMO

We have previously identified antimony (Sb) as a newly nerve poison which leads to neuronal apoptosis. However, the relationship between Sb exposure and Alzheimer's disease (AD) process lacks direct evidence. HE staining and Nissl staining showed significant nerve damage after Sb exposure. Therefore, we further evaluated Sb-associated AD risk by detecting accumulation of ß-amyloid protein (Aß) and neurofibrillary tangles (NFTs) in the brains of mice exposed to Sb for 4 and 8 weeks, and even 1 year. The results showed that dose of 20 mg/kg induced Aß accumulation, but not tau hyperphosphorylation after exposure for 4 week. Eight weeks later, both 10 and 20 mg/kg dramatically triggered Aß accumulation and increased tau phosphorylation at ser199. At the same time, 20 mg/kg could also increase tau phosphorylation at ser396 and number of NFTs. One years later, we found all of AD hallmarks detected in present study showed positive results in the brains of mice exposed to Sb at 10 and 20 mg/kg. In summary, our data provided direct evidence of Sb-associated AD risk, drawing more attention to Sb-triggered neurotoxicity.

RESUMO

Perception of facial identity and emotional expressions is fundamental to social interactions. Recently, interest in age associated changes in the processing of faces has grown rapidly. Due to the lack of older faces stimuli, most previous age-comparative studies only used young faces stimuli, which might cause own-age advantage. None of the existing Eastern face stimuli databases contain face images of different age groups (e.g. older adult faces). In this study, a database that comprises images of 110 Chinese young and older adults displaying eight facial emotional expressions (Neutral, Happiness, Anger, Disgust, Surprise, Fear, Content, and Sadness) was constructed. To validate this database, each image was rated on the basis of perceived facial expressions, perceived emotional intensity, and perceived age by two different age groups. Results have shown an overall 79.08% correct identification rate in the validation. Access to the freely available database can be requested by emailing the corresponding authors.
